    I used to do non-fiction selection at my old library and loved it. We'd take it very seriously and tried to get the right blend of items that reflect customer demand (what's popular in one library isn't necessarily popular in another); seasonal trends; current interest (eg books about maths at the moment are popular due to the Du Sautoy programmes), and generally off the wall "different" things that you think people might like.
    Please stay safe in the sun and learn the A-E of melanoma: A = asymmetry, B = irregular borders, C= different colours, D= diameter, larger than 6mm, E = evolving, is your mole changing? Most moles are not cancerous, any doubts, please check next time you visit your GP.

    It's simple molecular biology, except i'm doing it with something slightly more complicated and shouldn't matter, except it's JUST NOT WORKING.

    At the risk of being seen as some kind of nice people stalker (I promise I'm not, I'm just to shy to contribute much), I work with people who do a lot of pcr... is it real-time? what are your problems? I've only tried it a couple of times (with a lot of assistance), but my colleages are literally doing pcr just about every day. Feel free to pm me with more details if you want (or not if you don't, I won't be offended).

    Real-time is easy.
    I'm trying to amplify bisulfite-treated DNA to determine methylation status and I can get one set of primers to work but not the other set. There is a trick I'm missing but I'm not sure what. If any of that makes sense, I can PM with more details.

    They don't do so much real-time which was why I asked ;)
    I think that they do something (or are planning to do so) regarding methylation status. I might have mis-remembered though.

    Feel free to pm me, chances are that they won't be able to help with your problems but you never know!

    Just to explain the comment about being a nice-people stalker... I've:
    1. 'rolled an easter egg'
    2. worked on the census
    3. seen fc's pop-up shop
    4. was offered some daily mail numbers from last weekend
    and I
    5. work in science
    I think thats about it :o

    I've lurked on the house prices board since before it became 'hidden' :o

    Hi Nikkster :hello: Don't be shy......If you contribute more then I can add to my repertoire of things I have learnt on this thread that I have zero real life experience of........I may learn more about PCR's to go with small holding info, feral chickens, teens with learning difficulties, how librarians select books and help their customers, how to move to 2 (or was it 3?) houses within 4 weeks, F1, sheep, caring for ill parents.....and all the other things that go on here.

    Makes me feel more of a rounded human being rather than being trapped in my little bubble world sometimes.

    No, hammered silver. It's lovely :) And small ... and simple. Like me: small and simple.

    I did a practice piece in bronze first - and will be making some bronze ones when I get my own hammer at home.

    Sounds like a great way to spend a week.......and brave of you too to spend a week with strangers. I would struggle with a week like that myself and am not speshul at all. Did you eat some fruit or fresh veg today?


    Liking the holiday tales too SS and MS....was thinking about you both today as the rain chucked it down in London for hours on end.

    We have the trade show this weekend and OMG I am wired to to the max........the entire range just didn't climb out of my head in time (by that I mean a fully factory ready pattern and sealed sample ready to go) but some did.
    Dreading being 'on display' for 3 days to all the trade...it feels like being naked in public.

    It amazes me how quickly you turn this stuff round FC - I would imagine it would take weeks to put one garment together. I? am not a naturist but despite my pot belly I have no hang ups about being seen naked whereas for DW it is a huge deal, strange how we all have different things that worry us.

    We had the rain last night, today was sun and cloud with a cool breeze - poor kids were freezing during their swimming lesson, the Med has a lot to be said for it. Tomorrow we are going to a theme park near Totnes - being MSE I have found a money off voucher. I am feeling uncomfortable about leaving the builders so long and am really starting to feel uncomfortable at my parents house - just the being in someone else's house thing so am quite ready to go home next Tuesday (if I can stick it out till then).

    Citibank are I think FSA compensated and do a EUR account.

    They're all lovely - and we're all of the same mind re the tutor being .... chaotic/annoying/disorganised... and they have even been timing how long it takes to extract some metal out of him (most are experiencing 1-1.5 hour delays between asking for 6" of any metal and actually getting it in their hand :)

    Today's diet was:
    Sausages, eggs, beans, toast, coffee, jam (that's fruit, right!)
    2 pastries (you get those with morning/afternoon coffee)
    3 chocolate giant cookies (purchased Tuesday night)
    Pie/chips.

    :)
    it feels like being naked in public.
    Luckily I don't know how that feels.....
