Yes this is a possibility, as DWP calls are often from 0800 numbers.

If you're expecting a call that's been confirmed you will know what time it's been booked. The DWP use multiple 0800 number's so again if you managed to block them they'll just be rerouted. Probably also check your blocked number list against (Who Called Me) check

I took the appointment phone call today at 4:00pm and it went fine, but a strange thing happened where another DWP number tried to call me beforehand and the phone marked it as a missed call immediately.
The missed call happened (no opportunity to answer) at 3:55pm on an 8000 number. I didn't call back because I didn't know it was a DWP number and I didn't want to miss the appointment at 4:00pm.
The appointment call happened at 4:00pm on an 0800 number, which I answered and had the chat, and everything was fine.
Should I enquire on the journal as to the 3:55pm 8000 number call, or just leave it be?
EDIT:
I'm thinking that the DWP wanted to see if my phone number was still valid, so they tested it with the 8000 missed call, found it existed, and called me at 4:00pm, five minutes later. Simple as that.

Just leave. They probably tried to call you, realised it was 5 minutes early and phoned again.

You will get a message in your journal. So you know to expect a call.

I will be going abroad this June, to Cyprus, on a week-long trip paid for by my sister, but I'm wondering how I should tell the DWP and how it will affect my UC. Do I simply state the dates of the trip? Do I have to put it somewhere in particular? Will they reduce my UC for that time?

It depends on what you have agreed in your commitments.
Have you agreed to commitments that require you to be available and looking for work, or are you in a regime where you have been found unfit for work?

I'm not expected to look for work, but I heard I could be fined if I don't at least tell them when I'm going.

It will only affect your UC if you are out of the country for more than 1 month, so a 1 week trip abroad will not affect your UC. You can notify them of the date you depart and your expected return date.

You notify the period of your trip by reporting a change of circumstances in your UC account. There is an option there to record Going outside of GB (or an option with a similar title).
