Sign it electronically, an e-signature , if you are submitting by email ( no need to print or sign unless sending or delivering it on paper, usually its email only these days )2

Ideally you submit it on a weekday between 9 and 5, as Dolly sang, when they are working ( I wouldn't be sending it at 1 am personally )
Check that you used the correct address, send it again if necessary, after checking your spam folder as well as your inbox ( it's probably in your spam folder and you didn't look )
BTW , The 12 steps say, if you are printing and signing it, not that you must print it , but email is always the preferred method for the last few years since the pandemic
THE FIRST 12 STEPS:- Print your Defence if you are physically signing it.
- Sign it and date it.
- Scan the signed document back in and save it as a pdf, OR if you have no access to a scanner/printer then put an electronic signature in.
- Send your signed & dated pdf as an email attachment to ClaimResponses.CNBC@justice.gov.uk (the second one in the image above) but you MUST get an acknowledgement straight back or it is NOT submitted.
- Just put the claim number (check it very carefully) and the word Defence in the email title, and in the body of the email something like 'URGENT - CLAIM XXXXXXXX - Defence attached.
- IMPORTANT - MAKE SURE YOU GET AN EMAIL ACKNOWLEDGEMENT BACK FROM THE CNBC! After filing your Defence, there is more to do.
2 - Print your Defence if you are physically signing it.

Its an instant response, not a delayed response, if you didn't receive it within a few minutes, it's not coming, so your defence, may not have arrived , they are closed late afternoon and tomorrow, and Sunday too
Feel free to beat around the bush, manana, but personally i would be following the plan of action i outlined, not yours, especially as they close at 5pm and wont reopen until Monday at 8am, so nothing can be done on a weekend
You snooze, you lose ( a dangerous game you are playing here ) , but its your case, your problem, I would personally get it sorted out this morning3 -

Which email provider are you using? Just a couple of days ago another poster was having the same 'non-delivery' issue. They were using a talk-talk email address. Switched to another email account and it went through - and acknowledged - immediately.Please note, we are not a legal advice forum. I personally don't get involved in critiquing court case Defences/Witness Statements, so unable to help on that front. Please don't ask. .
