It could be a genuine mistake, and you'd probably just let it go without a second thought if they'd paid promptly in the first place.
I know it's the principle you're concerned about rather than the amount, but it would seem a bit mean and petty to ask them to pay the 5p owed. A better approach imo would be to economise on packaging (e.g. used Jiffy bag rather than a brand new one), or if the p&p price was for 1st class, downgrade to 2nd. The buyer probably won't even notice if you do, but if you want them to be aware that you're making a point, you could enclose a cheque for the difference between the 5p and postal downgrade.
