Do I have to leave the £500 in the account for a certain amount of time for it to qualify for the interest rate?
2k in each account, transferred 500 into first account then as soon as it showed transferred it to the next, and now going to move it to my ISA. Is that ok?

Cross fund the TSB accounts, ie £500 TSB1 to TSB2, and £500 back again. If these balances will be static, ie pure savings accounts, you can automate these monthly transfers with a pair of standing orders.
And 3 standing orders will completely automate things...
£506.50 from 1 to 2
£500.00 from 2 to 1
£13.00 from 2 out to another savings account held elsewhere.
NB: £6.50 is the average interest earned per month after savings tax deducted.0 -
