I plan on submitting this tomorrow now to the CCBCAQ@Justice.gov.uk email address from the guidance on the suggested defence template.
Out of interest, in my defence I made some references to the Leasehold agreement of my girlfriend. Do I need to submit the leasehold along with the defence in this first instance, otherwise how can they validate my points there? Or do I definitely just keep that for the WS and that evidence at that stage?

No - defence opens up the legal arguments, witness statement elaborates and provides evidence.

My defence has now been sent off! Let's see how this all pans out.
I will next work on drafting my witness statement, my girlfriend's witness statement along with that respective evidence to attach to them1 -

I did @Jenni_Dand have filed that in my emails. I have just checked on the MCOL and it says my defence has been received in the Claim History too

Double check the list of things to do in the guide to court. There is more to do at various stages before you submit the WS and exhibits.
Also, just because the court business centre has received your defence in a timely manner, you won't know how long it will be before they send it on to the claimant, and therefore won't know their deadline for deciding whether or not to continue.I married my cousin. Out of interest the Directions Questionnaire from the CCBC, should I fill that out and send it to them pre-emptively or wait until they send one to me and then send it back in reply?

Wait until your MCOL history says one has been sent out, and then download and complete a copy.
