Installing Windows 10 from usb ISO problems

Hi, I hope you can help.

Trying to upgrade vista to Windows 10. I have done the following:

Downloaded 64bit iso from Microsoft website (not via the tool as not supported)
Downloaded the 64bit version
downloaded rufus version 2.17 (latest version will not run on vista)
Followed a video regarding which settings to use
did the conversion - all seemed ok
restarted pc and I just get a blinking curser on top left of screen

I have checked that the bios is set for usb boot - it is (which is evidenced by the blinking screen)

I have tried to google the problem but cannot find a solution. Some say to change certain settings but they are not available so unsure what to do next.

My specs are Intel core 2 duo E8400 3.00GHz
6GB RAM
currently running 64bit vista SP2

USB stick is a TDK 8GB

Rufus settings for ISO:
MBR partition for BIOS
File system : NTFS

Any ideas?
