My guess is the seller has hit on a scheme to extract a good wedge of extra cash out of unsuspecting buyers.0

If you bought something for £2,000 in a shop you’d expect a receipt. Why would you hand that amount to a total stranger and not ask for one?0

I lost a note while counting the money in my car, so I told him to double check the £2480 was all there. He said it was about £150 short so I recounted the money and it most certainly wasn't. He then counted it again and discovered I was only £20 short and ...

How difficult is it to count 125 £20 notes, anyway?
Here's 5 x £20 in a pile. That's £100.
Here's another pile of 5 x £20 = 2 x £100.
...
5 x £100 piles = £500.
5 x £500 piles = £2,500.

Looks like the OP never learnt his lesson in getting solid paperwork at point of sale, i.e. have all parties sign paperwork that agree the deal is complete at point of sale.

The first big mistake was paying cash - a banker's draft is the same as cash - no mistake there.
It isnt.
A bankers draft can be forged or stolen.
It is not an absolute guarantee of the money.
I'd transfer the money direct to his account though if possible. 95% of people have internet banking.
Hindsight of course is 20/20 but with the initial "confusion" over monies paid, i'd definitely have got a receipt.
TBF its got to the point whereby dealing with the general public for cars (either buying or selling) has got that much of a minefield its easy to see why people either trade in or sell to WBAC.0
