I'd do this, and you could get pretty decent tyres, fully fitted and balanced for around £60. I'm not sure where you were getting your £500 from OP.

That very much depends on the car... on my A3, anything less than £400 and you're into brands I've never heard of and wouldn't trust at 70mph (officer) on a Lake District dual carriageway in February.

Just checked the spare tyre and it's a new bridgestone tyre that came with the car. Should I stick that on the front and put the low tread tyre in the boot?

Why not, if it's a normal wheel and not too old.

No, that very much depends on the recommended size of tyre and I was basing my posts on what the OP stated was his!

Yes, that would be advisable (assuming it's a full size spare).

    Why one the back?

    From Michelin
Better control in emergency braking or tight corners when the roads are slippery.
Less risk of losing control of your vehicle, especially on wet surfaces
Better road holding, particularly in difficult situations, whether your car is front or rear wheel drive

Costco will insist on new tyre(s) being fitted to the rear.

Depends how old it is. An unused spare tyre will harden and perish over the years to a point where it's unadvisable to use it even if it is fully treaded.

If the spare is too old it might be worth buying two new tyres, fitting them to the car and the worst of your legal tyres becomes the new spare tyre. Throw away the old tyre and the spare.

If spare is in good condition then yes by all means fit it to the car.
Changing the world, one sarcastic comment at a time.

The car is from 2006 so the spare tyre 10 years old.
