Yes, the money does need to be in the account.
We'll use a TSB Plus Current account as an example.
Their current account offers 5% AER on balances of up to £2000.
So deposit £2000 and let it earn it's interest.
The important bit to note is that this account requires you to deposit £500 every month for you to earn the interest. So, transfer £500 in (either manually or via standing order) and then transfer the £500 out (can be done on the same day, in the same hour; even the same minute).
