No it doesn't need to be connected to a computer to do that. I would restart the phone once you ticked USB debugging. Believe me it will solve the problem you are experiencing

The battery does better after a couple of weeks when the battery has been conditioned properly. Remember to run down the battery completely before you charge it for the first few weeks. If you are having problems with battery life then I would recommend looking at how often your apps are updating - for example does the likes of Facebook need to be updating every hour in the background? I generally switch background updates off and it significantly improves battery life too. You may also want to download an app called Juicedefender Beta which disables internet while the screen is off etc and makes the battery last longer. Also remember that things like Live Wallpapers are going to eat battery life too.

Please do not do a factory reset as you will still have exactly the same problem one you have wiped it.
