On 29th August, I received an email from DCB Legal with an attached N180 Directions Questionnaire completed by them. However, when I logged into MCOL, I couldn’t see any update on my case file. Should I wait for the update to appear, or can I download the N180 form myself and send it in? Thanks.
This is already explained in the first 8 steps.

Thanks. Submitted N180 and now received a mediation appointment for 6th Nov.

Should I prepare myself to explain to the mediator my defence? Any clues as to what questions will be asked?

Why are you planning on doing that instead of doing what the 8 steps tell you to say?

    Sure, thanks.

The mediation call is with regards to PCN Ref1 and I am still waiting for a claim to be raised against me with regards to PCN Ref2. During this mediation call, can I mention Ref2 also and try to settle both?

I doubt it,  especially if DCB Legal havent raised a claim against you

But you aren't trying to 'settle' a scam.

you just say zero offer and thats it , you the end the call
literally 2 minutes on the phone
I do these 3 to 4 times a week now.  So you can take this as fact

i dont wish to waste any time, my offer to the claimant is zero, they will discontinue - i will expect your email saying the claim will continue and mediation was not successful
