I wasn't sure if there was the same delivery number on labels bought at PO, but the fact that the OPs receipt mentioned it had delivery conformation suggests that PO labels also have it.

It's very hard to understand how the OP's sister posted this large letter.
Considering your post earlier re you buying your labels online so you get delivery confirmation even for 2nd class large letter, I doubt the OPs sister bought the label online so I don't think she will have tracking/delivery confirmation for the service she bought.

You may also need your delivery postcode. Then you will find out whether rm think they have delivered it, know they haven't, or aren't sure. If they think it's delivered it may be hard to claim it's lost.
