If my wife's money, Premium bond value and investments are inherited by me are they still subject to estate value calculation and inheritance tax. Do I then get the full allowance from her when I am deceased or do I only get the excess from her allowance from £325k

Anything left to a spouse or civil partner is exempt and does not use any of the NRB, so your estate would be entitled to the full transferable value, unless she has made gifts in the preceding 7 years or left other assets to others.0
