Since April 6th is a Saturday do we have to wait until the 8th to open this year's ISA?

And, as ever, it's the funding date that matters, rather than the opening date....

Whoever has the best rates on that day as I am after a 5 year fixed rate.

Whoever has the best rate on the Saturday is still likely to hold that status on the Monday, but you could always open one during the week (or more) before if you saw a rate you wanted to secure.

I am also hoping to open a 5 year Isa with monthly interest for 24/25 tax year, the best I can find so far is Paragon at 4%, does anyone know of a best one.
the other alternative, giving the new rules, is to open a 1 or 2 year one at a higher rate with say 10k then put just 10k in the 5 year one,d

Unlikely IMHO - there used to be an identifiable 'ISA season' like that but things moved on.  For example, looking through archived copies of the MSE ISA page, the leading five year rates on 12 April 2023 were exactly the same products and rates as on 2 April....
