I use USA Amazon site
to purchase and send things to people who live in the USA and am paying for USA Amazon Prime, so that deliveries are free and quick. I also use the UK Amazon site for
purchasing things for myself here in the UK and am also paying for UK Amazon Prime
for the same reasons as mentioned. Is it
necessary to pay for both UK and USA Amazon Prime, or will the UK one be
accepted for USA Amazon purchases?

One Amazon account will usually work on all the international Amazon sites.
However Prime is usually on a per country/region account, so buying Prime in the UK doesn't give you Prime in the US for example.
On occasion Amazon will run a deal with free international shipping with prime membership but that's only for a temporary period.
