Flight delay of two hours leading to missing car hire slot at Malaga Airport

Delay resulted in missing slot for car hire pick up. Costs incurred of about £300 - car hire and taxis from Airport to accommodations and back to airport.
Possible claim against Ryanair or Holiday Insurance?

Definitely no claim against the airline.  Check insurance but I think that is unlikely.  I've never heard of fixed time slots to collect a vehicle as flight delays are commonplace. When I've booked cars for collection at airports they generally request flight numbers so will be aware of the delay.   Who was the car hire company?

Most cars I've hired you've had to give an ETA for collection but the only real issue happens when your evening flight is delayed to the extent that the car hire place has closed for the night by the time you get there. If its £150 each way to your accommodation by taxi I'd be much more tempted to stay at the airport hotel for the night, collect the car in the morning and go check in at your original accommodation (obv inform them on the night so its not cancelled as a no-show).
