Can anyone explain the best way to set up a remote desktop between 2 machines over the internet running Windows 10 and both locations are using Sky routers?

    Which versions of W10? (Home / Pro).

    What's the end goal?

    If they(or one of them) are W10 pro use RDP , if not look at VNCviewer/server(or TightVNC), you could also use Teamviewer but I gave up on that since they assume I am using it in a none personal way and block me.

    TeamViewer is pretty good, but it's only free for private non-commercial use, and apparently they are cracking down tightly on this. 
    A kind word lasts a minute, a skelped erse is sair for a day.

    Easiest solution is TeamViewer.  Nothing to configure, just stick it on both machines and you can either feed them the Teamviewer Partner IDs or add them to a Teamviewer account that is signed in on both computers.  TightVNC/UltraVNC/TigerVNC are other options, though these sort of programs aren't particularly secure according to the likes of Kaspersky and CVEDetails, and they need ports to be opened in the router.

    Alternatively there is this, which is Free for up to 10 computers, bit more work involved to get it going but once you've done it, it just works.  No router configuration needed.

    I want to be able to access my Dad's computer to help with a few issues he's having but he's in isolation. He's W10 home, I'm W10 pro.

    Anydesk is also good - free for private use, and no checking as Teamviewer does, although this has been relaxed in the current situation, have switched to Anydesk from Teamviewer..

    Anydesk even works on my Raspberry Pi, Teamviewer can as well..

    Team viewer

    had an Indian Scammer wanted me to install it last week

    Told me I was having computer problems and he could fix it

    So it must work!!

    (needless to say I did not give the scammer the code just wasted his time for an hour :) )

    Are you a TalkTalk customer?  They block Teamviewer by default and you have to log in to your account on their website and disable all the "security" features to make it work again.

    I use it to support my parents because trying to get a sensible explanation of what they can see on the screen when I tell them to do something is hard.  But I had to go to their house (120 miles each way) to get their account reconfigured to allow me to remote in using Teamviewer.

    With regards to Teamviewer, or any other 3rd party software for that matter, would I still be required to set up (or rather try and get my dad to) port forwarding on his sky router?
