It will take time. It works in the same way as an electric kettle - just a heating element immersed in water.

If the water still doesn't get hot then maybe the immersion heater element has failed.

The thermostat strapped to the side of the cylinder should control when the boiler needs to provide heat - within the hot water time limits set by the time controller.

How long have you left it on? It takes a fair while to heat the whole tank, but if it is working you will be able to hear it 'fizzing'.

If there's no hot water after 30-45mins the element has probably gone, they don't last forever, we've got through 2 in the last 14 years. The connection would still be live, just the element won't heat anymore. It's not an expensive job to it replaced.

However, that doesn't explain why the boiler isn't heating the water. Do you have a control panel/programmer for the boiler?

I think its time to get professional in to be honest.
It could be a couple of things impossible to diagnose over the interwebs.
When you switch the hot water on at the timer is the boiler firing up?

If it is check for a bleed port and bleed air out of tank note not all tanks have this.

If the boiler not firing up when hot water is switched on. Then there a few things that could be at fault

    1.Programmer not calling for heat
2.thermostat on tank not calling for heat.
    3.stuck diver value
4.boiler not accepting call for tank heat.
    5.some dodgy wiring
