If the often stated reason for nitrogen that "the oxygen is smaller and leaks out quicker" really made a difference, then yes. Each time you topped up you'd be increasing the overall % of nitrogen present.
In practice, as others have pointed out, any variation using air is going to be more because of poorly seated beads or valves (which nitrogen will make no difference to) and / or water vapour which a well set-up compressor with moisture traps will prevent and, in any case, won't make enough difference for any normal driver to worry about.0
