Can't copy a large file to USB stick - Help?

I am trying to copy a back up .tib Maxblast file to a brand new Lexar 64GB USB Stick. But it's not happening.... I know that things always hold less than stated and it says on the properties 58.4GB free space the file I'm copying is 49.3GB but when I paste I just keep getting the message "The disk in the destination drive…

Thanks - yes I have the main back up on an external HDD this is my extra copy!
