My PC has been infected - A demand for £100 and PC locked. I cannot even get to Safe mode by pressing F8.

Will completely reloading everything cure the problem?

If so how do I go about this please?

Win7 32bit, running McAffee

If you search around the net there are ways to be able to get past this nasty bit of ransom ware, though it has been getting nastier so that you can not circumvent it.
Probably the best thing is to do a fresh install of Windows using the supplied Windows discs or if you have a machine with Windows pre-installed choose the boot device to be the back-up system install. The second method will vary from PC to PC. So best to consult the manual.

Have burnt that disk on wife's PC but I cannot open my CD draw. I get a white screen but can hear the PC windows opening sound and Skype starting.

Will a full re-install of Windows clear the bug?

Yes, but its more of a last resort measure (unless losing all your installed programs and having to backup all your data is no great hardship)

Can you open the CD drive manually? Some models have a teensy weensy little hole (like thin paperclip wire size) which you can use to manually force the door open.

What happens say, if you pay the £100?

I'm not suggesting for a minute do it, I'm just wondering if the !!!!!!!os that infect your computer unlock it for you?

I had this recently, what to do is head over to bleeping computer forums and there's a section on there for spyware removal where someone will help you out - reinstall is the very last resort!

There's different variants that achieve this is different ways so there's no "one size fits all" approach (other than backup/reinstall) to clean it.

Unplug from the internet and see if it will start in any of the safe modes: Safe mode, SM with networking, SM with command prompt (The latter may prove to be the only option).

If you can start explorer (SM with command prompt), you can navigate the file system and run Malwarebytes if you have it, or something fom a USB drive.

Otherwise you could try a USB based rescue disk if you're having problems with CD/DVD's

Bleeping offer excellent support, but I can do this here if the OP wants.

I used that on a friends laptop with the same issue (and I now know more about his browsing history than I really wanted to!!!).
It took a couple of hours (think I set it to scan aboslutely everything).

    It did work though.

I backed that scan up with the Kaspersky version just to make sure.

You may or may not get your files back - but for certain you will encourage more trouble, and will get yourself on an 'easy mark' list for the next load of malware going.
