It could be a the audio drivers. Check to see if the audio drivers have microphones enabled and that it isn't set to mute.
If this is fine, double check the microphone settings on the task bar and on Skype.
Then check to see if the microphone cable is connected to the correct port. If it's a USB headset, ignore the previous statement.
If all else fails, then it's likely to be a fault with the headphones.0
