I realise I was being slightly vague in my original post, let me explain a bit of background.
We have visited the USA a fair bit and done multi centre stays, always sorting our own flights.
Pretty much always used Kayak or Opodo or similar to book the 3 or 4 legs we needed.
This time however, I have been pondering on simply getting a return trip to one of the major carriers hub airports, then sorting ourselves out with ' internal ' flights.
As for where exactly we want to fly from / to..... well its early days. Toying with getting to JFK or Philly or Atlanta then onto Nashville , San Diego, Vegas, Miami, open to ideas really, but thats another forum.
I will take a look individually at the airlines you nice people have mentioned above and see what I can come up with.

Another one to remember is that some US airports have low taxes, fees and charges and thus spending frequent flier scheme miles can be good on one way flights in the US, especially those that you get 'free' with credit cards, blindman's tip btw.Posts are not advice and must not be relied upon.0

I tried to book cheap US internal flights earlier this year and found them prohibitively expensive. In the end I went to Trailfinders, it was the best thing I ever did as the man there kindly told me with so many different flights required (8 in total) I should also be on 'one ticket' in case anything went wrong.
If you make lots of separate flight bookings with individual tickets, one carrier will not care if you miss a connection with another flight because of delay/cancellation/schedule changes etc. However, most airlines have formed alliances and if you stay on 'one ticket' (mine is BMI/United Airlines/United Express) then it is their responsibility to make alternative arrangements if things change outwith your control.
Adding internal US flights onto my main UK>US flight didn't work out that much more. For example, Edinburgh>London>Denver only went up £20 when I added >Rapid City(return) on the end, even though the cheapest one-way internal flight I could find was $90. I now have three internal flights (on different days) throughout my holiday and it was actually still cheaper than the best price I found online for booking everything separately. Since I booked, one connection has had two minor time changes, but TF just send me a new notification, and once I'm in the US it will be United's job to rebook me if it's a major change. Fingers crossed UA don't get into a mess with the Continental merger tho...

I would second pins132 with that advice. Since I replied earlier we've started pricing up a winter trip to both the east and west coasts of the USA: flying into NYC and out of SFO, with a domestic flight in between. Searching with and without the internal flight between east and west coasts only brings up a difference of about £60 or £70, which is better than any of the standalone fares I can find on Virgin America etc.
If you're planning a multicity trip (A -> B, B -> C, C -> A) try searching for simple roundtrips A -> B and and A -> C with as much flexibility as the booking engines allow. Find the cheapest outbound and return dates, and then start searching multicity options. Most booking engines won't give you the usual +/- 3 days flexibility when searching multicity itineraries, probably because they're more complex to execute in the software.0
