I agree with Jem8472 - It could just be a speck of black dirt trapped underneath the stone that shows up when you look through the stone.
Take it to be cleaned and see how it turns out. Its surprising how much rubbish gets caught in the holey bits around the stones.0 -

Sounds like a bit of dirt under the setting to me. If so it should come out with ultrasonic cleaning at the jewellers.0

or you could just try strong alochol, traditionaly a lot of people used gin and a soft childs tooth brush.

I have been into the jeweller's today and they have confirmed the black spot is an inclusion. I can't understand how we never noticed it before, and my husband said he checked the diamond thorouhly before he bought it - the shop certainly never pointed anything out to him. Never mind, I shall just have to ignore it! Thanks for the advice everyone.0
