Yousendit is great! Although there are limitations IE:25 downloads of a file, if the recipient doesn't get the full file first time then they have to use up another one of the 25 chances. Obviously once those 25 have chances of downloading have been used then you have to upload the file again for others who missed it. Whereas Megaupload holds the file indefinately (unless it hasn't been downloaded for 30 days) and the link is re-useable (useful if the link is being put on a forum). Depending on what you want to do with your file and/or it's size will determine which service is best for you.
