Retiring from the Appointment of Trustee of a Discretionary Trust

I would like to retire from an appointment of a Discretionary Trust before the Deed of Trust has been created. I would like to know how to do this. At the moment there are two trustees that have been appointed who are uncle and niece. The niece wishes to go against the Letter of Wishes but not all of it. There is a clause in the will that says if anyone challenges the will they receive nothing. I am a residual beneficiary of the Estate and a Trustee of the Trust, also a beneficiary of the Trust at the bottom of the order of people who benefit from that Trust. I would really appreciate your help. I am set to inherit nothing even though I have been very good to my Mum, the only person who has which it states in her letter at the end of the WIll. Thank you
