Paying off part of card

Spent some money last month, let's say £10,000 on a credit card.
A bill has been produced and a DD will take the amount from a specific bank account in about 7 days.
If I "make a payment" to the card and pay off half, I guess the DD will only claim the remaining £5,000?
It's an American Express credit card.

Possibly not. You maybe too close to the DD date for it to be altered1

I'll just pay the money into the specific bank account and let it all happen automatically.
Just want to maximise it being in my Chase account before transferring it to an account that has zero interest.0
