It usually doesn't matter how many transfers are required, as long as the monthly pay in requirement is met by the end of the month in question. It's entirely possible to pay in £625 twice to qualify for the requirement (eg. pay in, transfer it out, then pay it back in again immediately). You could even break it down further if you needed (eg. 5x £250, 30x ~£42, or even 1250 x £1...)0

If you have a joint account with your wife then it's open to interpretation as to whether "you" includes her also. If you don't have a joint account then I think it's a stretch to include her as being "you" simply because you are married so I would think it's okay.
To avoid any confusion it would be better to be from a non Nat West account, something like a simple savings account would suffice.1
