Two questions;
1. Yes, but Google and look at your Local Authority's website for "HMO" ( HOUSES IN MULTIPLE OCCUPATION) for your added responsibilities, including the likely obligation that your Council will impose for you to register with them (mine does).
2. Yes, but again, use Google and look at the Planning Portal and your Local Authority's website for what you can do without Planning permission ("Permitted Development") and what they are likely to agree with PP. My Council even publish handy online guidance to " Residential Extensions...etc" with sketches and plans of preferred side, rear, loft and basement extensions.
I once chucked up a partition wall ( metal or wooden "stud work " topped with plaster board and finished with skirtings) to turn a through-lounge back into the two rooms which would originally have been there in a late Victorian house. An easy DIY job which didn't need any permissions.
But, while we didn't need Planning Permission to convert a garage into an additional habitable room in our current home, we did involve the Council "Building Control" staff. They checked that our builder did it right; regulatory stuff like foundations, thermal insulation and fire safety. Worth doing right, as their sign off certificate will be needed if ever we sell. But they did put a note on a national and local register of Council Tax bandings that in future, we might have to get up-rated and pay more!1 -
Both are possible, but both have potential hoops to jump through, so you need to do research.

Many BTL mortgages don't permit renting by the room so be careful choosing your lender. Ditto landlord insurance.0
