My cooling fan has recently started cutting in (noisily) every ten minutes or so when I am playing games. I have checked that the vents are not blocked. Does this suggest that my machine is on its way out?

Not necessarily - is this a new game or one that you've played for some time?
I assume you're talking about case, power supply or graphics card fans and not the CPU fan that should be running all the time?
If it's a temperature controlled case fan then something
is blocking airflow through the case...
Anyway, what I would do is to open the case and carefully hover out the dust and fluff that will have found its way into it. It may be worth cleaning the heatsinks with a bit of compressed air, too.0
