Can anyone advise me as to what I need to do regarding the Land Registry and Title Deeds etc.
I have paid off my mortgage and have been advised by Santander that they've advised the land registry to remove the charge on my property, but do I need to do anything? Do I need to contact the Land Registry? Do I need to acquire Title Deeds etc?
Many thanks for any advice/help.

No.
Assuming the LR remove the Charge, your electronic Title document at the LR will be updated. You should get a copy in due course showing this.
You can always check yourself by paying £3.00 and downloading a copy of your Title from them here.
It is just possible that your mortgage lender holds other historic documents relating to your property, though since it sems your property is Registered, these oo would likey be held electronically by the LR.
If Santander do hold any relevant paper copies, they will return them to you.
Give it time, then check the process has worked.0
