Block up airvent or not? Advise please

With gas cookers, unless the room is VERY small, no need for an air vent, as long as there is either a door or window that opens directly to outside air, i.e. not into a conservatory etc. However, there may be good reasons for the vent to be there, don't block it till you have discussed with your Gas Safe engineer at your annual boiler service. As has been suggested, it is possible to block the vent from outside if needed, even by fixing a weather proof board over it, so no need to do it immediately.0
