iOS 10 not updating

I want to update my iPhone 6s Plus through iTunes on Win7.
I always do it this way, as there can be problems doing it over the air.
To update to 10.0.1, I have to update to iTunes 12.5.1.
No matter how many times I press the download iTunes button, it won't update.
Any idea how to force it to update?
I have never had a problem like this before.

I've had problems in the past with updating iTunes via Wi-Fi. Turning off the Wi-Fi and reconnecting my laptop to the router with an Ethernet cable has always worked for me.
Check your task bar in case there is a window waiting to be opened to say ok to the upgrade.
I should add that iTunes is very bad at showing you what it's doing and how far it has progressed with an update so it may be doing it in the background and you just have to be patient. I have a traffic flow reporter on my router so I can see the download activity via the wired connection which is handy....
