If I was to use my M&S Credit Card to pay my Barclaycard bill would this be classed as a cash transaction and theresubject to cash interest?
I am asking, as I am trying to use my M&S Card for my main spending in order to accumulate points.

Normally this would count as a balance transfer, attract a fee and interest, and gain no points.
There are offers from time to time that remove some of these drawbacks. Whether they apply in your case I don't know.0
