Sick computer

So I wake up this morning to be greeted by the wife telling me she has broken the computer!
Not sure what she was on last night but when I turned it on I was getting the infected Metropolitan Police scam website that it kicked into once the computer was turned on.
Not a problem as I have had this before so I press f8 when booting up so I can do rstrui.exe as I have done previously. Problem is that this doesn't seem to work now - having done Restore with Command Prompt the computer rather than go to my Windows homepage goes to Shutting Down page.
Rather more concerning when I try a couple more times to re-boot it is coming up in a grey box Location is not available C;\windows\system32\config\systemprofile\desktop refres to a file that is unavailable.
Is this one I can sort myself or do I need to take it to a professional??
Any advices greatly appreciated,

    Once you've fixed it (see above if ti's the same one)

    Create a separate Administrator account to enable you to more easily deal with issues like these .

    Many thanks for the replies guys.
    However, seem to be falling at the first hurdle! I've clicked on the link given, however when I go to download HitmanPro it goes to the webpage and has a line saying download HimanPro 3.7 but no link to click on ??
    Any advice

    Thanks NiftyDigits - have managed to download HitmanPro onto my flash drive and am now running it on my infected PC - fingers crossed this will have the desired effect.

    Out of interest whats a good free anti-virus program? After todays scare I'm thinking I best have a clean up of my desktop and laptop.

    I use Avast, others may well suggest Avira, AVG (wouldnt go near it myself, Avast has been fine so why change it) or another 1. If you have a Barclays bank account you can get a paid anti virus for free, its Kaspersky if its useful.
