Glass blocks are very heavy - a whole wall of them would probably need support from below.

A curtain as a door would be an option for a temporary doorway If you intend to return the room to its original state after a few years then I don't see what all the fuss is about really. We are living in times when up sticks and moving just isn't the option that it was 10yrs ago sadly.
