Getting a car which has a usb port to connect your memory stick so that the car audio can play the mp3 music from the memory stick.
How do you set up the mp3 music on the memory stick?
1. have mp3 files at top level
2. create several folders containing (for example) 20 mp3 songs each
or
3. as the handbook suggests create a .m3u format playlist.
If it is done by option 3 how does one create a .m3u format playlist?

1 and 2 will depend on the capabilities of the software the car stereo uses - 1 will always work, 2 should work if the stereo is reasonably good (I suspect any new player will cope with that, but there are always exceptions to the rule).
The main advantage of 2, is that it's easier to organise the music by artist/album (or even both), which means you can normally then set the player to just play that folder.
You can use a number of free programmes to create an m3u playlist on the PC, the obvious one is the faithful old Winamp
I'd guess you copy the music over to the memory stick, open up Winamp (or whatever program you use), load the music from the memory stick into the programe, create a playlist and then save it to the memory stick.
Personally I tend not to bother with playlists most of the time on my PC, or mp3 player, and instead just set the players to random0
