Just to clarify, can I put my spare 250GB SATA 5400rpm HD into my PS3?

I know you can change the hard drive to make it bigger, which is what I want to do...

It needs to be a laptop hard drive (and 9.5mm thick if I recall)
If you put it in, it simply reformats and you start afresh. If you wish to backup your current drive, you need an EXTERNAL drive formatted to FAT 32 and backup first:idea:0 -
RIK is spot on. I would make a backup of your PS3 in its current form first, whip the hard disk out and install the new one.
From personal experience I would look at removing the disk when the PS3 is cold i.e. hasn't been turned on for a while. I found the screws were less receptive to being taken out when the unit was warm and almost managed to balls it up.
Apparently hard disk swapping does not invalidate your warranty either so no worries there!

No, after inserting the new drive into the PS3 and powering on it will take you through the steps to get it all working. Take a look at this link, a pretty comprehensive step by step process on how to do it from start to finish!
