Absolute beginner at spreadsheets with another simple question!

Sorry about this but can anyone help?
How do i make it so column C automatically subtracts B and puts the result in E?

In cell E1 put =C1-B1

Now copy the contents of cell E1 into the cells below (E2, E3, etc. as far as you need).

Or if you have lots of cells to put it into highlight Column E from the first cell with the calculation to the last and press Ctrl + D, or go to edit --> Fill --> Down. This will insert the calculation into all cells highlighted below the first.
