Be careful about stacking too much on the passenger seat - you must be able to see out of the passenger window, and you must have a clear view of the wing mirror ! You don't want to get pulled over by the police before you ev en get there ;-)
Keep your tarpaulin beside you on the passenger seat, and get that down straight away before you open the boot, to stake your claim on floor space. But buyers will walk all over it, so bear that in mind if you plan to display things that need to be kept clean such as clothing.
Then wait for a few minutes for the vultures to move on to the next new arrival before you start unpacking the car - they'll come back if they're relly interested.
And if they start rooting in the boot of your car as you're unloading, I find that a loud verbal warning along the lines of "I'm closing the boot now - please move your hands before they get trapped" then following through with the action has very good effects ... ;-)
If you can get hold of the shallow boxes that the supermarkets use to display bananas, you can just lift them straight out and take the lids off if you're selling things like books.
I'm planning on selling at a few car boots this year, and I use boards and folding trestles rather than pasting tables. I've borrowed a rear-mounted cycle carrier to transport the trestles, as they take up far too much space in the back of the car, and I can get them set up before I open the boot, then just slide the boards off the top of the boxes.
If you are able to, park with the passenger side of your car along the length of your selling space - you gain a bit more space, and you don't have to turn your back on your items when you get things out of the boot.
You can then open the window on the passenger side of the car, and things you may want to keep within reach but not on display can stay on the passenger seat, out of sight of buyers but accessible by you.
And don't forget to take an extra platic sheet to cover everything in case it decides to rain :-(
