If you want to find the volume without being scientific, take a 1 litre measuring jug, fill it up to 500ml with water, place the Tube of cream in the water (push it just under surface with finger if necessary) and read the water level. Well that sounds very scientific to me!
However, in this scenario then you are not really taking into the account the mass of the tube itself. It could be a tube with really thick walls and just a teeny weeny bit of cream inside it!0 -

Security is not going to take it out of the ziploc bag and check if it is under 100mls.
They know what looks more or less like 100mls and from what you say, this is well under.
In any case, as someone else has said, take a photocopy of the prescription (front and back), or get a fresh prescription before you go and take that with you, and you'll have no problems as you are allowed to take on board medication that is essential to have with you on the flight.
I know it isn't the same, but I have to take medical equipment in my hand luggage and it doesn't meet the requirements of all the low frills airlines, but when I explain they always let me through (touch wood!)I'm a retired employment solicitor.
