My partner has acquired a car from his brother and he has posted him the new keeper suppliment for the car and my partner has insured the car in his name. Unfortunately the M O T has been lost and the brother seems to believe that he taxed another car once before in the main Post office without the M O T because they checked there and then. As the car isn't yet in my partners name he can't tax this on line I believe? Or is the only way out to get duplicate at an M O T station for £10?

You can check the MOT status at http://www.motinfo.gov.uk with the registration number and the doc ref from the V5 / V5 new keepers slip.0

Get the brother to tax the car online, give him the money. Just like buying a car that has tax on it already.
That's assuming the DVLA have sent him the tax reminder.Member of the first Mortgage Free in 3 challenge, no.19

The PO dont have access to the MOT info site, which is why you need to present the MOT at th PO.
