You don't have to, but you can if you like.

If you choose to put in £500 on 1st June, you'll get an extra £3.42 gross in interest compared to leaving it at the twelve payments that you've already made.

I opened same account last july my annivarsary date will be the 10 of july.
I have made 11 deposits starting from the opening date in july 2008 to may 2009. i think the next payment in june will make it 12 deposits therefore i wont need to make another deposit by july.

Also i want to know the best time to move the money after the annivasery date as i have other things lined up to use the money

You don't actually move the money from the Regular Saver yourself, Halifax do it for you as stated in your terms and conditions -

Maturity of term - after 12 months your savings and interest are simply transferred into your nominated savings account (which you'll be asked to open at the same time as opening your Regular Saver). Your Regular Saver account remains open and you can continue to save - the interest rate applicable at the time is then fixed for another year.

Once in your nominated account the money is yours to move around as you wish. Do remember that the Regular Saver isn't closed and the Standing Order will start another year's saving going (at the interest rate current when the first deposit is received, currently 4%) unless you cancel it.

You can if you like though ... it'll increase your total interest by £1.37 gross if you do make a 13th payment of £500 on 1st July!
