Yes, it almost certainly will, because the overall amount of heat you're asking the boiler to generate is lower, which in turn means less gas needs to be burned.
There are some exceptions to this, depending on the type of house you're in, but it's a good idea for most people.
You can check the effect it's having very easily: Read your gas meter and note the reading down. Don't make any changes to your radiator settings - just carry on as normal for a week. Read the meter again, and work out how many units you've used. Now make your changes - turn down (or better, turn off) radiators in unused rooms. Do this for a week and read your meter again - hopefully you'll have burned less gas in the second week than the first.

That will only give you a realistic comparison if the outside temperatures are the same both weeks. However , as was said, if you are asking your system to produce less heat then it will use less gas. So don't wait for a week, adjust your thermostats now!0
