Adding a second hard drive?

Sorry for an ignorant question...I have a 3 year old desk top with 80g hard drive which is getting full, i have a 250g external for backing up it and others.
am i right in thinking that i can add a second hard drive inside the desktop to create more storage, whilst all the programs still run from the existing c-drive?
is it an esy job that gets recognised when plugged in and there seem to be several types of hard drive styles - how do i find which one is compatible?
