Do you nean 2016:17 through to 2019:20?
What exactly do you mean by "my Self Assessment shows we have made a claim"?
Are you referring to your Self Assessment calculation for each tax year? Have you compared those to the previous calculations?
Have you checked your Self Assessment account to see if the credit (presumably) now due is shown?
Remember Marriage Allowance doesn't always result in a refund, for 2019:20 it might reduce the tax owed and being collected via your current tax code in which case you would get a new tax code for 2021:22.1 -

My wifes tax code has obviously been amemend for current and past years hence she has ended up with a tax bill of over £400 in total for all years being claimed
My Self assessment shows
A refund of £250 was received by me for year 20/212019 to 2020 Applied for Marriage Allowance retrospectively 2018 to 2019 Applied for Marriage Allowance retrospectively 2017 to 2018 Applied for Marriage Allowance retrospectively 2016 to 2017 Applied for Marriage Allowance retrospectively

A tax code can't be amended for tax years that have ended. Do you mean she has had calculation showing tax now owed?
If your Self Assessment account does show "Applied for Marriage Allowance retrospectively" then you wouldn't be due any refund, as your application would cancel our the benefit of your wife applying.
If your wife is the only person who has applied then have you checked your Self Assessment statement of account?

So your wife's tax office have actioned her request but your tax office has not yet actioned the transfer for previous years yet.
Current years are dealt with first and the previous years are dealt with later.
HMRC did have large backlog or post to be actioned due to processing returns from April and the dealing with the covid grant applications.
Did you ask when the claim might be dealt with?
