Marriage tax allowance, claim now or after 05/04/21?

If I am eligible to claim marriage tax allowance, does it matter if I claim this tax year or next tax year? If I claim after 05/04/21 for 2020/21's allowance, will I get a cheque for £250 instead?

If you are claiming marriage allowance transfer, it would be your spouse who would be eligible for any refund.
However, assuming you mean your spouse is claiming to transfer 10% to you....it's so late on that it doesn't matter when you claim, it will be dealt with under the year end calculation as there isn't time to issue a coding notice to your employer.You keep using that word. Whoever the recipient is it is too late now to get any refund via PAYE.
If a valid application is made now for 2020:21 the recipient should get a calculation from HMRC in the summer and be invited to pull the refund (c£248 if no other tax overpaid or owed) into their bank account via their Personal Tax Account.
A cheque would only be sent if they ignored the request to do a bank transfer.1 -

You would only get one if your application results in you having some tax to pay (or you have overpaid for some other reason).

If you are entitled to claim for earlier years as well, you lose the ability to claim for one year every time 5 April passes.1
