You can't actually "claim" Marriage Allowance.
Your spouse/civil partner had to apply.
They can do this without a NINO, it is covered on gov.uk.0 -
I know, I mean is there anything I need to do in order to make sure it can be backdated. Any documents I need etc.0

Which tax year is the first one your spouse/civil partner wants to apply for?0

Ideally last year and this year. Hopefully later this year she will be able to get an NI number, but the backlog is massive so who knows...0

She has until 5 April 2024 to make an application for the 2019:20 tax year.
Not that leaving it right to the last minute would be advisable but no real issue waiting for the NINO, you would just get a bigger backdated refund if she cannot apply until into the next tax year.
The benefit to you for 2019:20 and 2020:21, assuming you are eligible and can get the full benefit, is likely to be a tax refund of c£495.2 -
