I would agree with everyone above that they just want to discuss how they intend to process the unpaid leave.
When we have had similar situations with new employees, we will give them a few options of how to treat it, mainly so we don't leave them with a very low first months wage and possibly leave them struggling. If someone has a large number of unpaid days, we can sometimes split the deduction over a number of pay periods.

OP - I don't think I've ever encountered anyone who has told a potential employer that they have a holiday booked, they offer them the job and then say they can't take the holiday. Don't worry, I'm sure it'll be fine.0
